Then you would need transillumination and the kind of vertical support a microscope offers. I take it you are employing an infinity-corrected objective with a tube lens in between?

Most microscope objectives will require a cover glass and from 80X onwards, most need oil or other fluid for immersion as well. I wouldn't mess with such stuff on any focusing rail, rather use the microscope which is designed for the purpose.

 

Metallurgical objectives generally are designed without requirement of cover glass and have longer working distances, but still high magnifications are better handled by a microscope setup. I decided not to extend my line of Mitutoyo objectives beyond the 20X class.
